Welcome to circRNADisease v3.0

Circular RNAs (circRNAs) are a major class of covalently closed non-coding RNAs with emerging roles in disease initiation, progression, therapeutic response, and clinical outcome. By systematically reviewing more than 18,000 published articles, circRNADisease v3.0 currently documents 15,390 manually curated circRNA–disease entries involving 7,774 circRNAs, 677 MONDO-standardized diseases, and 66 species. In addition, circRNADisease v3.0 integrates 7,159,865 mutation–circRNA associations across 30 TCGA cancer types and 2,962,504 copy number variation (CNV)–circRNA associations across 33 TCGA cancer types.

Compared with the previous release, circRNADisease v3.0 includes several major improvements: (i) a more than two-fold increase in circRNA–disease entries, together with substantially expanded circRNA, disease term, disease cross-reference database, and species coverage; (ii) expanded entry-level annotations for circRNA–disease entries, including genomic location, subcellular localization, validation methods, clinical significance, data availability, and code availability, which were not available in v2.0; (iii) a newly added confidence score for each circRNA–disease entry, calculated based on clinical evidence, circRNA identity verification, expression detection methods, validation methods, and other experimental support; (iv) a new Circ2CNV module for retrieving and visualizing copy number alterations within circRNA loci across TCGA cancers; and (v) a new Circ2Celltype module for querying cell-type-specific circRNAs and exploring their reported expression patterns in defined cell types.
