circRNA-disease information
Expression pattern:
DN
Associated gene: CREBZF, ADAR1, STAT3
Associated microRNA: miR-1269a
Biological function: inhibits proliferation; promotes apoptosis; suppresses tumor growth
Molecular mechanism: ADAR1 binds flanking intron complementary sequences to suppress circPAPD4 circularization; circPAPD4 sponges miR-1269a to increase CREBZF; CREBZF suppresses STAT3 dimerization and ADAR1, forming a positive feedback loop (circPAPD4/miR-1269a/CREBZF/STAT3/ADAR1).
Biological pathway or process:

proliferation (inhibits); apoptosis (promotes); ceRNA regulation (other); JAK/STAT (inhibits)

Detected method:
Q
H
M
Validation methods:

Microarray; Bioinformatics Analysis; RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; Nuclear-Cytoplasmic Fractionation; FISH / smFISH; Sanger Sequencing; ISH (In Situ Hybridization); Clinical Sample Validation; Survival Analysis; Pearson correlation analysis; RNA Pull-Down; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); Transfection; CCK8; EdU Staining; Annexin V/PI Flow Cytometry; TUNEL; Western Blot; Co-IP; In Vivo Animal Model; IF (Immunofluorescence); H&E Staining

Clinical significance:

Low circPAPD4 expression was substantially correlated with larger tumor size, advanced tumor-node-metastasis (TNM) stages, and higher Ki-67 expression of BC tumors; patients with lower circPAPD4 expression presented an unfavorable RFS; circPAPD4 expression served as an independent prognostic factor for RFS in BC.

Description:

circPAPD4 is down-regulated in breast cancer and functions as a tumor suppressor by inhibiting proliferation and promoting apoptosis. It mainly localizes to the cytoplasm and acts as a ceRNA by sponging miR-1269a to increase CREBZF, which inhibits STAT3 dimerization to reduce ADAR1 and thereby enhances circPAPD4 biogenesis in a positive feedback loop.

Confidence score:

0.8941
