circRNA-disease information
Expression pattern:
UP
Associated gene: WNT2B
Associated microRNA: miR-424-5p
Biological function: Circ_0000064 promotes high glucose-induced HRMC proliferation, cell cycle progression, inflammatory responses and extracellular matrix accumulation; circ_0000064 knockdown attenuates these diabetic nephropathy-related cell dysfunctions.
Molecular mechanism: Circ_0000064 acts as a miR-424-5p sponge to relieve miR-424-5p-mediated inhibition of WNT2B, thereby regulating HG-induced mesangial cell dysfunctions.
Biological pathway or process:

proliferation (promotes); cell cycle (promotes); inflammation (promotes); fibrosis (promotes); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; RNase R Treatment; Clinical Sample Validation; Transfection; CCK8; EdU Staining; Cell Cycle Assay; ELISA; Western Blot; Bioinformatics Analysis; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation)

Clinical significance:

Circ_0000064 was upregulated in kidney biopsy samples from DN patients and was proposed as a biomarker in DN management.

Description:

In this study, circ_0000064 was upregulated in HG-treated human renal mesangial cells and in kidney biopsy samples from DN patients. Functionally, circ_0000064 promoted HG-induced proliferation, inflammation, cell cycle progression and ECM accumulation, while its knockdown alleviated these DN-related phenotypes. Mechanistically, circ_0000064 acted as a miR-424-5p sponge to upregulate WNT2B, supporting the circ_0000064/miR-424-5p/WNT2B axis as a potential biomarker or therapeutic mechanism in DN.

Confidence score:

0.7591
