Associated microRNA: -
Biological function: circSRPK1 promotes the malignant phenotype of gastric cancer, including proliferation, tumorigenesis, migration, invasion and metastasis, and inhibits apoptosis.
Molecular mechanism: circSRPK1 is regulated by MAGOH/EIF4A3 and directly interacts with hnRNP A2B1 to promote hnRNP A2B1 nuclear translocation and binding to the ESE element on RON mRNA exon 7, thereby regulating RON mRNA alternative splicing, inducing RONdelta160 production, and promoting GC progression.
Biological pathway or process:

proliferation (promotes); apoptosis (inhibits); migration (promotes); invasion (promotes); metastasis (promotes); other pathway/process (promotes)

Detected method:

H
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; Actinomycin D / DRB Stability Assay; RT-qPCR; FISH / smFISH; ISH (In Situ Hybridization); Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; RIP (RNA Immunoprecipitation); RNA Pull-Down; IF (Immunofluorescence); Transfection; CCK8; EdU Staining; Colony Formation Assay; Transwell Assay; Wound Healing Assay; In Vivo Animal Model; H&E Staining; IHC (Immunohistochemistry); Western Blot; Cohort Study; Survival Analysis; Bioinformatics Analysis

Clinical significance:

High circSRPK1 expression is associated with adverse outcomes, advanced tumor stage, higher lymphatic metastasis rate, and decreased overall survival in GC patients; circSRPK1 is proposed as a promising biomarker and target for GC prognosis and treatment.

Description:

circSRPK1 is an SRPK1-derived human circRNA upregulated in gastric cancer tissues and cell lines. It promotes GC proliferation, tumorigenesis, migration, invasion and metastasis by interacting with hnRNP A2B1 to regulate RON mRNA alternative splicing and induce RONdelta160 formation, while its production is enhanced by the upstream MAGOH/EIF4A3 axis. High circSRPK1 expression is associated with advanced disease features and poor overall survival.
