circRNA-disease information
Expression pattern:
UP
Associated gene: E2F2, Ago2, PD-L1, IFN-gamma, IL-10, CD8 + T cells, bax, bcl-2, cleaved-caspase-3, Ki-67
Associated microRNA: miR-637
Biological function: Promotes NSCLC cell proliferation and immune escape, inhibits apoptosis, and accelerates tumor growth in vivo; knockdown suppresses proliferation, promotes apoptosis, reduces immune escape markers, and suppresses xenograft tumor growth.
Molecular mechanism: circ_0000877 acts through a ceRNA-like mechanism by directly interacting with miR-637, leading to regulation of the downstream target E2F2; E2F2 overexpression reverses the effects of circ_0000877 knockdown.
Biological pathway or process:

proliferation (promotes); apoptosis (inhibits); immune regulation (promotes); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Cohort Study; Survival Analysis; Bioinformatics Analysis; Transfection; CCK8; EdU Staining; Annexin V/PI Flow Cytometry; Flow Cytometry(Non-apoptosis/cycle); ELISA; Western Blot; Luciferase Reporter Assay; RNA Pull-Down; RIP (RNA Immunoprecipitation); In Vivo Animal Model; IHC (Immunohistochemistry); H&E Staining; TUNEL

Clinical significance:

Up-regulated circ_0000877 was linked to tumor size, differentiation, TNM stages, and short overall survival, indicating poor prognosis; the study suggests circ_0000877 might be a potential biomarker for NSCLC diagnosis.

Description:

In this study, circ_0000877 was up-regulated in NSCLC tissues and cell lines, and high expression was associated with clinicopathological progression and poor overall survival. Functionally, circ_0000877 promoted NSCLC cell proliferation, immune escape, and tumor growth while suppressing apoptosis. Mechanistically, circ_0000877 directly interacted with miR-637 and regulated the downstream target E2F2, forming a circ_0000877/miR-637/E2F2 axis.

Confidence score:

0.8067
