circRNA-disease information
Expression pattern:
UP
Associated gene: EZH2, OCT4, Nanog, Ki67
Associated microRNA: miR-124-3p
Biological function: circ-TRPS1 promotes PCa proliferation, migration, invasion, metastasis, and cancer stem-like cell differentiation; circ-TRPS1 knockdown suppresses these malignant phenotypes.
Molecular mechanism: circ-TRPS1 functions as a miR-124-3p sponge to enhance EZH2 expression, thereby promoting EZH2-mediated stemness and PCa progression.
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); metastasis (promotes); stemness (promotes); ceRNA regulation (other); other pathway/process (promotes)

Detected method:

S
Validation methods:

RNase R Treatment; circRNA-seq; RT-qPCR; FISH / smFISH; Clinical Sample Validation; Bioinformatics Analysis; Luciferase Reporter Assay; RNA Pull-Down; Transfection; CCK8; Colony Formation Assay; Transwell Assay; In Vivo Animal Model; IHC (Immunohistochemistry); Western Blot

Clinical significance:

High circ-TRPS1 expression correlated with aggressive PCa phenotypes and unfavorable prognosis; circ-TRPS1 may be a prognostic/predictive biomarker and candidate therapeutic target.

Description:

circ-TRPS1 is up-regulated in high-grade prostate cancer tissues and PCa cell lines, and high expression is associated with aggressive clinicopathologic features and poor prognosis. Functionally, circ-TRPS1 promotes PCa proliferation, migration, invasion, metastasis, and cancer stem-like cell differentiation by sponging miR-124-3p and increasing EZH2 expression. Its knockdown suppresses tumor growth and metastasis in vitro and in vivo, supporting circ-TRPS1 as a potential therapeutic target.

Confidence score:

0.8439
