E2F7, Ago2, p21, Cyclin D1 |
| Associated microRNA: |
miR-545, miR-589 |
| Biological function: |
Promotes proliferation, migration, invasion and tumorigenesis of LAC cells; supports cell cycle progression; may reduce sensitivity to gefitinib inhibition. |
| Molecular mechanism: |
circPRKCI is overexpressed partly due to PRKCI/3q26.2 amplification and acts through a ceRNA mechanism by sponging miR-545 and miR-589, relieving suppression of E2F7 and altering p21/Cyclin D1 signaling. |

| Clinical significance: |
High circPRKCI expression correlates with larger tumor size, higher TNM stage and shorter overall survival; circPRKCI may serve as a potential biomarker and therapeutic target for LAC. |
| Description: |
circPRKCI is a PRKCI-derived circular RNA from the 3q26.2 amplicon that is up-regulated in lung adenocarcinoma. It promotes LAC cell proliferation, invasion/migration and tumor growth by sponging miR-545 and miR-589 to increase E2F7 and regulate downstream p21/Cyclin D1 signaling. High circPRKCI expression is associated with larger tumors, advanced TNM stage and poor overall survival, and targeting circPRKCI suppresses xenograft growth and enhances gefitinib inhibition in EGFR-mutant cells. |
