| Biological function: |
promotes cancer stemness (tumorsphere formation, tumorigenicity), invasion, tumor growth, and immune evasion; enhances resistance to PD1 blockade |
| Molecular mechanism: |
circFAT1 binds STAT3 in the cytoplasm, prevents SHP1-mediated dephosphorylation of STAT3, promotes STAT3 activation and inhibits STAT1-mediated transcription; induces CXCL9/CXCL10 changes and impacts CD8+ T cell infiltration |

| Clinical significance: |
High circFAT1 expression is associated with lymph node metastasis and poor prognosis (shorter overall survival) in HNSCC patients. |
| Description: |
circFAT1 is up-regulated in HNSCC and promotes tumor growth, invasion, cancer stemness, and immune evasion. It acts mainly in the cytoplasm by binding STAT3 and blocking SHP1-mediated STAT3 dephosphorylation, thereby sustaining STAT3 activation and suppressing STAT1-driven type I IFN transcription; circFAT1 knockdown increases CD8+ T-cell infiltration and improves anti-PD1 efficacy. |
