circRNA-disease information
Expression pattern:
DN
Associated gene: MOB1B, QKI, CELF4, FLNB pre-mRNA, YAP, TAZ
Associated microRNA: miR-3127-3p, miR-1915-3p
Biological function: represses CRC cell proliferation, invasion, migration and xenograft tumor growth; promotes apoptosis; acts as a tumor-suppressive circRNA; chemotherapy-induced circFLNB biogenesis contributes to tumor inhibition
Molecular mechanism: circFLNB acts as a miRNA sponge for miR-3127-3p, blocks miR-3127-3p-mediated MOB1B mRNA degradation, regulates the MOB1B/Hippo pathway, and is generated through chemotherapy-induced QKI- and CELF4-dependent alternative splicing of FLNB pre-mRNA
Biological pathway or process:

proliferation (inhibits); invasion (inhibits); migration (inhibits); apoptosis (promotes); Hippo/YAP (other); ceRNA regulation (promotes); other pathway/process (promotes)

Validation methods:

Microarray; RT-qPCR; RNase R Treatment; FISH / smFISH; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; Bioinformatics Analysis; Luciferase Reporter Assay; RNA Pull-Down; RIP (RNA Immunoprecipitation); Transfection; CCK8; EdU Staining; Annexin V/PI Flow Cytometry; Transwell Assay; Wound Healing Assay; In Vivo Animal Model; IHC (Immunohistochemistry); IF (Immunofluorescence); TUNEL; Western Blot; Cohort Study; Survival Analysis; RNA-seq

Clinical significance:

higher circFLNB expression is a potential biomarker for better prognosis in CRC patients; clinical models based on circFLNB can predict CRC prognosis and may guide clinical diagnosis and treatment; circFLNB may also serve as a chemotherapy response biomarker

Description:

circFLNB is a human FLNB-derived circRNA that is down-regulated in colorectal cancer but induced by chemotherapy. It suppresses CRC proliferation, migration, invasion, and xenograft growth while promoting apoptosis, mainly by sponging miR-3127-3p to upregulate MOB1B and modulate the Hippo pathway. Chemotherapy promotes circFLNB biogenesis through QKI- and CELF4-dependent alternative splicing, and higher circFLNB expression is associated with better prognosis.
