circRNA-disease information
Expression pattern:
UP
Associated gene: PD-L1, CDK4, CDK6
Associated microRNA: miR-34a
Biological function: promotes AML cell proliferation
Molecular mechanism: circ_POLA2 suppresses the maturation/production of mature miR-34a, leading to increased PD-L1 expression and enhanced proliferation.
Biological pathway or process:

proliferation (promotes); cell cycle (promotes); ceRNA regulation (other)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Survival Analysis; Transfection; CCK8; BrdU; Cell Cycle Assay; Western Blot; Bioinformatics Analysis

Clinical significance:

Patients with higher expression levels of circ_POLA2 had lower overall survival rate.

Description:

circ_POLA2 is upregulated in AML bone marrow samples and AML cell lines, and its high expression is associated with worse overall survival. Functionally, circ_POLA2 promotes AML cell proliferation, at least in part by suppressing the production/maturation of mature miR-34a and increasing PD-L1 expression.

Confidence score:

0.6458
