Associated gene: c-JUN mRNA, c-MYC mRNA, CDK6 mRNA
Associated microRNA: -
Biological function: inhibits cell viability and proliferation; promotes apoptosis; inhibits G0/G1 phase transition (suppresses cell-cycle progression) in arsenic-induced malignantly transformed lung cells
Molecular mechanism: circBRWD1 directly binds c-JUN/c-MYC/CDK6 mRNAs and reduces their mRNA stability, decreasing their expression (and protein levels), thereby restraining malignant transformation/proliferation under arsenic exposure
Biological pathway or process:

proliferation (inhibits); apoptosis (promotes); cell cycle (inhibits); mRNA stability (inhibits)

Detected method:

S
Validation methods:

RNA-seq; RT-qPCR; RNase R Treatment; divergent primers PCR; FISH / smFISH; Nuclear-Cytoplasmic Fractionation; Transfection; CCK8; EdU Staining; Annexin V/PI Flow Cytometry; Cell Cycle Assay; Luciferase Reporter Assay; RNA Pull-Down; Actinomycin D / DRB Stability Assay; Western Blot; Bioinformatics Analysis

Clinical significance:

circBRWD1 has potential as a diagnostic biomarker and therapeutic target for lung cancer

Description:

circBRWD1 is down-regulated in arsenic-exposed malignantly transformed bronchial epithelial cells and in multiple lung cancer cell lines. Functionally, circBRWD1 suppresses proliferation and cell-cycle progression and promotes apoptosis. Mechanistically, it directly binds c-JUN/c-MYC/CDK6 mRNAs and decreases their stability, reducing oncogenic mRNA/protein expression and thereby inhibiting arsenic-induced lung carcinogenesis.

Confidence score:

0.7958
