circRNA-disease information
Expression pattern:
UP
Associated gene: TNPO1, QKI, E2F7, Ago2
Associated microRNA: miR-27a-3p
Biological function: promotes proliferation; promotes migration; promotes invasion; promotes ferroptosis; promotes tumor growth and metastasis
Molecular mechanism: ceRNA sponging miR-27a-3p to upregulate TNPO1; QKI promotes circBCAR3 biogenesis via intronic binding; hypoxia-induced E2F7 transcriptionally activates QKI
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); ferroptosis (promotes); ceRNA regulation (promotes); metastasis (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; divergent primers PCR; FISH / smFISH; RIP (RNA Immunoprecipitation); Luciferase Reporter Assay; Bioinformatics Analysis; Transfection; CCK8; EdU Staining; Colony Formation Assay; Wound Healing Assay; Transwell Assay; Western Blot; Flow Cytometry(Non-apoptosis/cycle); In Vivo Animal Model; H&E Staining; IHC (Immunohistochemistry)

Clinical significance:

circBCAR3 has close association with tumor grading

Description:

circBCAR3 is upregulated in esophageal cancer and further induced by hypoxia. It promotes proliferation, migration, invasion, tumor growth/metastasis and enhances ferroptosis, mainly via a ceRNA mechanism (circBCAR3/miR-27a-3p/TNPO1); its biogenesis is promoted by QKI, which is transcriptionally activated by hypoxia-induced E2F7.

Confidence score:

0.8279
