Validation methods:

Back-Splice Junction PCR / divergent primers PCR; Sanger Sequencing; RT-qPCR; Clinical Sample Validation; Transfection; MTT; Transwell Assay; Cell Cycle Assay; Annexin V/PI Flow Cytometry; Western Blot; Bioinformatics Analysis

Clinical significance:

potential biomarker for the early diagnosis and treatment of HCC

Description:

hsa_circ_0002124 is up-regulated in HCC tissues and several HCC cell lines. Functional assays in 97H cells show that it promotes proliferation, migration and invasion, facilitates cell-cycle progression, and inhibits apoptosis. Mechanistically, it is proposed to act as a miRNA sponge and enhance MAPK pathway activity (p-JNK/JNK, p-ERK/ERK, p-P38/P38, c-Myc), suggesting biomarker potential for HCC.
