circRNA-disease information
Expression pattern:
DN
Associated gene: SIRT1
Associated microRNA: miR-145
Biological function: promotes INS-1 cell proliferation and insulin secretion under glucolipotoxicity and inhibits apoptosis, alleviating beta-cell dysfunction in T2DM context
Molecular mechanism: Acts as a ceRNA/miRNA sponge for miR-145 to relieve repression of SIRT1, thereby increasing SIRT1 expression
Biological pathway or process:

proliferation (promotes); apoptosis (inhibits); ceRNA regulation (promotes)

Detected method:
Q
M
Validation methods:

Microarray; RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; Nuclear-Cytoplasmic Fractionation; Transfection; CCK8; Annexin V/PI Flow Cytometry; ELISA; Western Blot; Luciferase Reporter Assay; Bioinformatics Analysis; Clinical Sample Validation

Clinical significance:

Potential novel diagnostic marker and therapeutic target for T2DM

Description:

hsa_circ_0115355 (also referred to as circCSE1L) is down-regulated in T2DM patient serum and in high-glucose-treated INS-1 cells. Functionally, it protects beta-cell models from glucolipotoxicity by promoting proliferation and insulin secretion and inhibiting apoptosis, mainly via a ceRNA mechanism in which it sponges miR-145 to upregulate SIRT1.

Confidence score:

0.7884
