| Biological function: |
inhibits melanoma cell migration and invasion and suppresses lung and liver metastasis |
| Molecular mechanism: |
circZNF609 binds the RBP FMRP and promotes FMRP binding to RAC1 mRNA, enhancing FMRP-mediated destabilization of RAC1 mRNA and reducing RAC1 expression |
| Biological pathway or process: |
migration (inhibits); invasion (inhibits); metastasis (inhibits); mRNA stability (inhibits) |

| Validation methods: |
Microarray; RT-qPCR; Back-Splice Junction PCR / divergent primers PCR; Sanger Sequencing; RNase R Treatment; Actinomycin D / DRB Stability Assay; Nuclear-Cytoplasmic Fractionation; FISH / smFISH; Transfection; Wound Healing Assay; Transwell Assay; In Vivo Animal Model; H&E Staining; RIP (RNA Immunoprecipitation); RNA Pull-Down; IF (Immunofluorescence); Western Blot; Bioinformatics Analysis; Survival Analysis; Clinical Sample Validation |
| Clinical significance: |
Low circZNF609 abundance is associated with distant metastasis, deeper Breslow depth, and shorter overall survival in cutaneous melanoma patients; circZNF609 is negatively correlated with Breslow depth and clinical stage. |
| Description: |
circZNF609 is down-regulated in acral and cutaneous melanoma and functions as a metastasis suppressor. It localizes mainly in the cytoplasm and binds the RBP FMRP to enhance FMRP-RAC1 mRNA interaction, decreasing RAC1 mRNA stability and RAC1 expression, thereby inhibiting melanoma migration/invasion and in vivo metastasis; low circZNF609 is associated with worse patient survival. |
