circRNA-disease information
Expression pattern:
UP
Associated gene: HMGA2, Ago2
Associated microRNA: miR-151-3p
Biological function: CircNUP98 promotes high glucose-induced proliferation, fibrosis, inflammatory response and oxidative stress in HMCs; its knockdown alleviates these injuries and increases apoptosis.
Molecular mechanism: circNUP98 acts as a ceRNA by sponging miR-151-3p, thereby positively regulating HMGA2; circNUP98 is also packaged into serum exosomes in DN.
Biological pathway or process:

proliferation (promotes); apoptosis (inhibits); fibrosis (promotes); inflammation (promotes); oxidative stress (promotes); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; Nuclear-Cytoplasmic Fractionation; Luciferase Reporter Assay; RNA Pull-Down; RIP (RNA Immunoprecipitation); Transfection; CCK8; Annexin V/PI Flow Cytometry; ELISA; Western Blot; ROC Analysis; Clinical Sample Validation

Clinical significance:

Exosomal circNUP98 shows diagnostic value for DN patients (AUC 0.9146).

Description:

circNUP98 is up-regulated in DN patient serum and in high glucose-stimulated HMCs. It mainly localizes to the cytoplasm and promotes HG-induced mesangial cell dysfunction (proliferation, fibrosis/ECM markers, inflammation, oxidative stress) by sponging miR-151-3p to upregulate HMGA2; it is also present in serum exosomes and shows diagnostic potential for DN.

Confidence score:

0.8455

Other information
Title:

Circular ribonucleic acid nucleoporin 98 knockdown alleviates high glucose-induced proliferation, fibrosis, inflammation and oxidative stress in human glomerular mesangial cells by regulating the microribonucleic acid-151-3p-high mobility group AT-hook 2 axis.
