circRNA-disease information
Expression pattern:
UP
Associated gene: AKT3
Associated microRNA: microRNA-124 (miR-124)
Biological function: Promotes ESCC cell proliferation, migration, EMT and tumor growth; knockdown suppresses these phenotypes.
Molecular mechanism: Acts as a miRNA sponge (ceRNA) for miR-124, thereby relieving repression of AKT3 and promoting ESCC progression.
Biological pathway or process:

proliferation (promotes); migration (promotes); EMT (promotes); ceRNA regulation (promotes); PI3K/AKT (other)

Detected method:
Q
H
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; RT-qPCR; ISH (In Situ Hybridization); Luciferase Reporter Assay; Transfection; CCK8; EdU Staining; Colony Formation Assay; Transwell Assay; Western Blot; In Vivo Animal Model; Clinical Sample Validation; Bioinformatics Analysis; Survival Analysis

Clinical significance:

High circHIPK3 expression in ESCC tumor tissues is associated with poor prognosis and correlates with lymph node metastases, greater tumor size, and poor differentiation.

Description:

circHIPK3 is upregulated in ESCC tissues and cell lines and its high expression is associated with poor prognosis and aggressive clinicopathologic features. Functionally, circHIPK3 promotes ESCC proliferation, migration and EMT, at least in part by sponging miR-124 to increase AKT3 expression, and its knockdown suppresses xenograft growth.

Confidence score:

0.8324
