circRNA-disease information
Expression pattern:
UP
Associated gene: RNF38, Smad2/3, p-Smad2/3
Associated microRNA: miR-449a
Biological function: circ_LRIG3 promotes HCC cell proliferation, migration, invasion and tumor growth, and inhibits apoptosis; circ_LRIG3 knockdown suppresses malignant HCC phenotypes in vitro and in vivo.
Molecular mechanism: circ_LRIG3 acts as a ceRNA by sponging miR-449a to positively regulate RNF38 expression, thereby promoting the Smad2/3 pathway in HCC.
Biological pathway or process:

TGF-beta/SMAD (promotes); proliferation (promotes); migration (promotes); invasion (promotes); metastasis (promotes); apoptosis (inhibits); EMT (promotes); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

RNase R Treatment; Actinomycin D / DRB Stability Assay; RT-qPCR; Clinical Sample Validation; Transfection; Colony Formation Assay; MTT; Annexin V/PI Flow Cytometry; Transwell Assay; Luciferase Reporter Assay; Western Blot; In Vivo Animal Model; Bioinformatics Analysis

Clinical significance:

Description:

circ_LRIG3 is up-regulated in HCC tissues and cell lines and functions as a tumor promoter. Its knockdown inhibits proliferation, migration, invasion, EMT-related marker changes, Smad2/3 pathway activation, and xenograft tumor growth while promoting apoptosis. Mechanistically, circ_LRIG3 sponges miR-449a to increase RNF38 expression and activate the miR-449a/RNF38/Smad2/3 axis.

Confidence score:

0.7595
