| Biological function: |
promotes chemotherapeutic resistance and cancer cell survival/growth under genotoxic (DNA damage) stress; inhibits apoptosis; represses R-loop accumulation |
| Molecular mechanism: |
p53 induces circASCC3 via transcriptional activation of ASCC3 and repression of SFPQ; circASCC3 binds DDX5 and prevents its proteasomal degradation, enhancing DDX5-mediated R-loop resolution |
| Biological pathway or process: |
chemoresistance (promotes); apoptosis (inhibits); other pathway/process (other) |

| Validation methods: |
Microarray; RT-qPCR; Sanger Sequencing; Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; Nuclear-Cytoplasmic Fractionation; Transfection; MTT; Annexin V/PI Flow Cytometry; Western Blot; RNA Pull-Down; RIP (RNA Immunoprecipitation); Luciferase Reporter Assay; ChIP / ChIP-seq; In Vivo Animal Model; IF (Immunofluorescence); Bioinformatics Analysis |
| Clinical significance: |
circASCC3 is lower in colorectal cancer tissues than matched adjacent normal tissues; higher tumor circASCC3 levels correlate with worse prognosis (not statistically significant). |
| Description: |
circASCC3 (hsa_circ_0077495), derived from ASCC3, is induced by p53 and mainly localizes in the cytoplasm. It promotes cancer cell survival and chemoresistance under genotoxic stress by binding and stabilizing DDX5, thereby enhancing R-loop resolution; SFPQ negatively regulates its circularization. In colorectal cancer tissues, circASCC3 is lower than in matched normal tissues, while higher tumor levels trend toward worse prognosis. |
