| Expression pattern: |
UP |
| Associated gene: |
SLC1A5, Ago2 |
| Associated microRNA: |
miR-330-3p |
| Biological function: |
Promotes NSCLC cell proliferation, invasion, EMT and glutamine metabolism; promotes tumor growth in vivo. |
| Molecular mechanism: |
Acts as a miRNA sponge for miR-330-3p to relieve repression of SLC1A5, thereby enhancing glutamine transport/metabolism and malignant phenotypes. |
| Biological pathway or process: |
proliferation (promotes); invasion (promotes); EMT (promotes); other pathway/process (promotes) |

| Validation methods: |
Bioinformatics Analysis; RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; FISH / smFISH; RIP (RNA Immunoprecipitation); Luciferase Reporter Assay; Transfection; CCK8; Transwell Assay; Western Blot; ELISA; In Vivo Animal Model; IHC (Immunohistochemistry); Clinical Sample Validation; Cohort Study; Survival Analysis; ROC Analysis |
| Clinical significance: |
High circ_0000518 expression was linked to bleak prognosis in NSCLC sufferers; ROC AUC 0.760 for distinguishing stage I patients from healthy controls; associated with tumor size, TNM stage, invasion depth and lymph node metastasis. |
| Description: |
hsa_circ_0000518 (circ_0000518), derived from the RPPH1 gene, is up-regulated in NSCLC tissues and cell lines and mainly localizes to the cytoplasm. It promotes malignant phenotypes (proliferation, invasion/EMT) and glutamine metabolism by sponging miR-330-3p to upregulate the glutamine transporter SLC1A5, and its knockdown suppresses xenograft tumor growth. Clinically, higher circ_0000518 expression is associated with worse survival and shows moderate diagnostic value (AUC 0.760). |
