circRNA-disease information
Expression pattern:
UP
Associated gene: SERPINH1 mRNA, SERPINH1, c-Myc, SRSF10
Associated microRNA: -
Biological function: Promotes NPC cell proliferation, migration, invasion, tumor growth and metastasis.
Molecular mechanism: circCAMSAP1 binds the SERPINH1 mRNA 3’UTR to increase SERPINH1 mRNA stability and expression; SERPINH1 binds c-Myc to inhibit its ubiquitination-degradation, increasing c-Myc; c-Myc (with SRSF10) promotes CAMSAP1 transcription and back-splicing to enhance circCAMSAP1 production (positive feedback loop).
Biological pathway or process:

mRNA stability (promotes); ubiquitination (inhibits); proliferation (promotes); migration (promotes); invasion (promotes); metastasis (promotes)

Validation methods:

RNA-seq; RT-qPCR; ISH (In Situ Hybridization); FISH / smFISH; Nuclear-Cytoplasmic Fractionation; RNase R Treatment; Actinomycin D / DRB Stability Assay; Transfection; MTT; Cell Cycle Assay; Wound Healing Assay; Transwell Assay; In Vivo Animal Model; H&E Staining; IHC (Immunohistochemistry); Western Blot; circRIP; Luciferase Reporter Assay; RNA Pull-Down; RIP (RNA Immunoprecipitation); Co-IP; IF (Immunofluorescence); ChIP / ChIP-seq; Bioinformatics Analysis

Clinical significance:

circCAMSAP1 expression was significantly correlated with high T stage, N stage, and M stage classification in NPC patients; suggested as a prognostic biomarker or therapeutic target.

Description:

circCAMSAP1 is up-regulated in nasopharyngeal carcinoma and promotes proliferation and metastasis. Mechanistically, circCAMSAP1 binds the SERPINH1 mRNA 3’UTR to enhance SERPINH1 mRNA stability and expression; SERPINH1 then stabilizes c-Myc by inhibiting its ubiquitination-degradation, while c-Myc (with SRSF10) further increases circCAMSAP1 biogenesis, forming a positive feedback loop.

Confidence score:

0.8489
