circRNA-disease information
Expression pattern:
UP
Associated gene: FABP3
Associated microRNA: -
Biological function: Desensitizes LUAD cells to ferroptosis by reducing lipid peroxidation; reduces AA incorporation into the plasma membrane and global AA; promotes NAT generation via taurine reaction, thereby suppressing ferroptosis.
Molecular mechanism: Exosomal cir93 elevates intracellular cir93; cir93 directly interacts with FABP3 and upregulates FABP3; FABP3 transports AA and facilitates AA reaction with taurine to generate NAT; NAT downregulates ACSL4/LPCAT3/PLTP via TLX2-dependent transcriptional regulation, reducing AA membrane incorporation and lipid peroxidation.
Biological pathway or process:

ferroptosis (inhibits); apoptosis (other); lipid metabolism (other); mitochondrial function (other); other pathway/process (other)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; FISH / smFISH; RNA Pull-Down; RIP (RNA Immunoprecipitation); CLIP; Transfection; Western Blot; In Vivo Animal Model; CCK8; Flow Cytometry(Non-apoptosis/cycle); Survival Analysis; Bioinformatics Analysis

Clinical significance:

High levels of cir93 predicted ferroptosis resistance and shorter overall survival in LUAD patients.

Description:

In LUAD, exosomal circRNA_101093 (cir93) is upregulated and maintains elevated intracellular cir93. cir93 binds to and increases FABP3, reducing AA availability and AA membrane incorporation (partly via NAT generation), which suppresses lipid peroxidation and desensitizes LUAD cells to ferroptosis; high cir93 is associated with poorer overall survival.

Confidence score:

0.7932
