circRNA-disease information
Expression pattern:
UP
Associated gene: RCC2, YY1, AGO2
Associated microRNA: miR-330-3p
Biological function: promotes HCC cell proliferation and tumor growth
Molecular mechanism: ceRNA sponge of miR-330-3p to upregulate RCC2; YY1 transcriptionally promotes circ-0015004
Biological pathway or process:

proliferation (promotes); ceRNA regulation (promotes)

Detected method:
Q
M
Validation methods:

Microarray; Bioinformatics Analysis; RT-qPCR; RNase R Treatment; Nuclear-Cytoplasmic Fractionation; Transfection; CCK8; Colony Formation Assay; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); ChIP / ChIP-seq; Western Blot; In Vivo Animal Model; Clinical Sample Validation; Survival Analysis

Clinical significance:

Higher circ-0015004 levels were associated with shorter overall survival, larger tumor size and higher TNM stage in HCC patients.

Description:

circ-0015004 (from SDHC) is up-regulated in HCC tissues and cell lines, mainly localized in the cytoplasm, and promotes HCC proliferation and xenograft tumor growth. It acts as a ceRNA that sponges miR-330-3p to increase RCC2 expression, and its transcription is positively regulated by YY1. High circ-0015004 expression is associated with worse clinicopathological features and poorer overall survival.

Confidence score:

0.8389
