| Expression pattern: |
UP |
| Associated gene: |
ERbeta, CXCR4, TMX4 |
| Associated microRNA: |
miR-622, miR-1303, miR-888 |
| Biological function: |
promotes lung cancer/NSCLC cell invasion and metastasis/progression |
| Molecular mechanism: |
ERbeta transcriptionally upregulates circ-TMX4 via binding TMX4 promoter (ERE3/4); circ-TMX4 sponges miR-622 to relieve repression of CXCR4 (3'UTR), increasing CXCR4 translation |
| Biological pathway or process: |
invasion (promotes); metastasis (promotes); ceRNA regulation (promotes) |

| Validation methods: |
RT-qPCR; RNase R Treatment; divergent primers PCR; Transfection; RNA Pull-Down; Luciferase Reporter Assay; ChIP / ChIP-seq; Western Blot; Transwell Assay; IHC (Immunohistochemistry); In Vivo Animal Model; Clinical Sample Validation; Bioinformatics Analysis |
| Clinical significance: |
circ-TMX4 is significantly elevated in tumor tissues and correlates with clinicopathological characteristics (tumor size, T grade, N grade, differentiation) |
| Description: |
In NSCLC/lung cancer, circ-TMX4 (hsa_circ_0001130) is up-regulated and promotes invasion and in vivo metastatic progression. Mechanistically, ERbeta binds the TMX4 promoter to increase circ-TMX4, which sponges miR-622 and thereby increases CXCR4 translation to enhance invasive behavior. |
