| Expression pattern: |

| Associated gene: |
HDAC4 |
| Associated microRNA: |
miR-29a-3p |
| Biological function: |
Promotes EC cell proliferation, cell cycle progression, migration, invasion, and tumor growth, and inhibits apoptosis; circ_FOXO3 knockdown suppresses EC progression in vitro and in vivo. |
| Molecular mechanism: |
circ_FOXO3 functions as a ceRNA/miRNA sponge for miR-29a-3p, thereby upregulating HDAC4 and promoting EC progression. |
| Biological pathway or process: |
ceRNA regulation (promotes); proliferation (promotes); migration (promotes); invasion (promotes); apoptosis (inhibits); cell cycle (promotes); metastasis (promotes) |

| Validation methods: |
RT-qPCR; Clinical Sample Validation; Transfection; CCK8; Annexin V/PI Flow Cytometry; Cell Cycle Assay; Transwell Assay; Luciferase Reporter Assay; Western Blot; In Vivo Animal Model; Survival Analysis; Cohort Study; Bioinformatics Analysis |
| Clinical significance: |
High circ_FOXO3 expression was associated with shorter overall survival, advanced FIGO stage, severer histological grade, and more lymphatic metastasis; circ_FOXO3 may be a promising therapeutic target in EC. |
| Description: |
circ_FOXO3 is up-regulated in human endometrial carcinoma tissues and cell lines and is associated with poor overall survival and aggressive clinicopathological features. Functionally, circ_FOXO3 promotes EC proliferation, migration, invasion, cell-cycle progression, and xenograft tumor growth while suppressing apoptosis. Mechanistically, it acts as a miR-29a-3p sponge to increase HDAC4 expression through the circ_FOXO3/miR-29a-3p/HDAC4 axis. |
