circRNA-disease information
Expression pattern:
UN
Associated gene: STAT3, PRKAA/AMPKalpha, IL6R, linear HIPK3 (linHIPK3), STK11
Associated microRNA: MIR124-3p
Biological function: circHIPK3 functions as an oncogene in lung cancer by promoting cancer cell proliferation, colony formation, migration and invasion, while suppressing autophagy in STK11 mutant lung cancer cells.
Molecular mechanism: circHIPK3 acts through a ceRNA-like MIR124-3p-STAT3-PRKAA/AMPKalpha signaling mechanism; circHIPK3 silencing releases MIR124-3p activity, downregulates STAT3, increases phospho-PRKAA/AMPKalpha, and induces autophagy in STK11 mutant lung cancer cells. circHIPK3 and linHIPK3 antagonistically regulate autophagy.
Biological pathway or process:

autophagy (inhibits); AMPK (inhibits); proliferation (promotes); migration (promotes); invasion (promotes); ceRNA regulation (other)

Detected method:
Q
Validation methods:

RNase R Treatment; RT-qPCR; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; Transfection; Colony Formation Assay; Transwell Assay; Western Blot; Survival Analysis; Bioinformatics Analysis

Clinical significance:

High circHIPK3:linHIPK3 C:L ratio was associated with poor survival, especially in advanced-stage NSCLC patients, and may serve as a prognostic factor; circHIPK3 alone did not show significant survival differences.

Description:

This study identifies circHIPK3 as an oncogenic circular RNA and autophagy regulator in NSCLC. circHIPK3 supports lung cancer cell proliferation, migration and invasion, while its knockdown induces autophagy through the MIR124-3p-STAT3-PRKAA/AMPKalpha pathway in STK11 mutant lung cancer cells. Clinically, the circHIPK3:linHIPK3 ratio, rather than circHIPK3 alone, was elevated in tumors and associated with poorer survival in advanced-stage NSCLC.

Confidence score:

0.7325
