circRNA-disease information
Expression pattern:
UP
Associated gene: hnRNPA1, HIF1A IRES, HIF-1 alpha, CD24
Associated microRNA: -
Biological function: circRSU1 promotes HCC malignant phenotypes, spheroid formation, colony formation, tumor growth, and enrichment of the CD24 positive cancer stem cell population.
Molecular mechanism: circRSU1 interacts with the RNA-binding protein hnRNPA1, increases hnRNPA1 protein stability by reducing proteasomal degradation, enhances hnRNPA1 binding to HIF1A IRES, boosts HIF-1 alpha protein translation, and increases CD24 positive cells.
Biological pathway or process:

HIF-1 signaling (promotes); stemness (promotes); proliferation (promotes); ubiquitination (inhibits); other pathway/process (promotes)

Detected method:
Q
S
M
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; circRNA-seq; RT-qPCR; Microarray; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; RIP (RNA Immunoprecipitation); Luciferase Reporter Assay; Transfection; Colony Formation Assay; Flow Cytometry(Non-apoptosis/cycle); In Vivo Animal Model; Western Blot; Cohort Study; Bioinformatics Analysis

Clinical significance:

circRSU1 is highly expressed in HCC tumor tissues and may serve as a potential therapeutic target for HCC patients enriched with CD24 positive CSC population.

Description:

circRSU1 is up-regulated in HCC tumor tissues and promotes malignant and stemness phenotypes in HCC cells and mouse models. Mechanistically, it binds hnRNPA1, stabilizes hnRNPA1 protein, enhances hnRNPA1 interaction with HIF1A IRES, increases HIF-1 alpha translation, and expands the CD24 positive CSC-like population.

Confidence score:

0.8676
