circRNA-disease information
Expression pattern:
DN
Associated gene: GLS / glutaminase
Associated microRNA: miR-27b-3p
Biological function: circSirt1 protects vascular smooth muscle cells against oxidative stress-induced injury, cell death and apoptosis during atherosclerosis, and promotes glutamine metabolism.
Molecular mechanism: circSirt1 functions as a miR-27b-3p sponge to relieve miR-27b-3p-mediated suppression of GLS, thereby restoring glutamine metabolism and protecting VSMCs from oxidative stress-induced injury.
Biological pathway or process:

apoptosis (inhibits); glutaminolysis (promotes); ceRNA regulation (promotes); other pathway/process (inhibits)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Bioinformatics Analysis; RNA Pull-Down; Luciferase Reporter Assay; Transfection; MTT; Annexin V/PI Flow Cytometry; Western Blot

Clinical significance:

circSirt1 may be a therapeutic target or approach for atherosclerosis treatment.

Description:

circSirt1 is down-regulated in VSMCs from atherosclerosis patients and under oxidative stress. Its overexpression protects AoSMCs/VSMCs from oxidative stress-induced injury and apoptosis by sponging miR-27b-3p, increasing GLS expression, and restoring glutamine metabolism. The circSirt1-miR-27b-3p-GLS axis is proposed as a potential therapeutic mechanism for maintaining VSMC homeostasis in atherosclerosis.

Confidence score:

0.6996
