circRNA-disease information
Expression pattern:
DN
Associated gene: ESR1, ERalpha, SMAD7
Associated microRNA: hsa-miR-96-5p
Biological function: hsa_circ_0004913 is described as a key regulator in the ERalpha/circRNA/miRNA/SMADs network; ESR1/ERalpha increases hsa_circ_0004913 expression, and hsa_circ_0004913 targets hsa-miR-96-5p within a network proposed to suppress HCC progression.
Molecular mechanism: ERalpha/ESR1 functions as a transcription factor binding the hsa_circ_0004913 promoter and enhances its expression; hsa_circ_0004913 interacts with hsa-miR-96-5p in a circRNA/miRNA/SMADs regulatory network involving SMAD7.
Biological pathway or process:

ceRNA regulation (other); TGF-beta/SMAD (other); proliferation (inhibits); migration (inhibits); cell cycle (other)

Detected method:
Q
M
Validation methods:

Microarray; RT-qPCR; Bioinformatics Analysis; Transfection; ChIP / ChIP-seq; Luciferase Reporter Assay

Clinical significance:

Description:

In this study, hsa_circ_0004913 was down-regulated in the HCC cell line MHCC97H compared with normal QSG7701 liver cells. ERalpha/ESR1 was shown to bind the hsa_circ_0004913 promoter and increase its expression, while hsa_circ_0004913 interacted with hsa-miR-96-5p within the proposed ERalpha/hsa_circ_0004913/hsa-miR-96-5p/SMAD7 regulatory axis. The authors propose that this circRNA/miRNA/SMADs network contributes to suppression of HCC progression.

Confidence score:
