circRNA-disease information
Expression pattern:
UP
Associated gene: HKDC1, AGO2
Associated microRNA: miR-3167, miR-192-3p
Biological function: Promotes CRC cell proliferation, migration/metastasis, EMT, glycolysis and xenograft tumor growth; inhibits apoptosis and oxidative phosphorylation; reduces chemosensitivity to oxaliplatin.
Molecular mechanism: circVMP1 acts as a ceRNA that sponges miR-3167, preventing miR-3167-mediated degradation of HKDC1 mRNA and thereby upregulating HKDC1 to activate glycolysis and promote CRC progression.
Biological pathway or process:

glycolysis (promotes); oxidative phosphorylation (inhibits); proliferation (promotes); migration (promotes); metastasis (promotes); EMT (promotes); apoptosis (inhibits); ceRNA regulation (promotes); drug resistance (promotes); chemoresistance (promotes)

Validation methods:

Sanger Sequencing; RT-qPCR; RNA-seq; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; RIP (RNA Immunoprecipitation); Luciferase Reporter Assay; Transfection; CCK8; Annexin V/PI Flow Cytometry; Transwell Assay; In Vivo Animal Model; Western Blot; IHC (Immunohistochemistry); Bioinformatics Analysis

Clinical significance:

circVMP1 might be a therapeutic target that would benefit CRC patients.

Description:

circVMP1 is upregulated in colorectal cancer tissues and functions as a tumor-promoting circRNA. It enhances proliferation, migration/metastasis, EMT, glycolysis and xenograft growth while suppressing apoptosis and reducing oxaliplatin chemosensitivity. Mechanistically, circVMP1 mainly acts through the circVMP1/miR-3167/HKDC1 ceRNA axis to increase HKDC1 expression and activate glycolysis.

Confidence score:

0.8252
