Reduced circDNAJC16 expression correlates with advanced pathological stage, lymph node metastasis, elevated serum CEA and poor prognosis; it may serve as an independent biomarker for predicting LUAD patient outcomes.
circDNAJC16 (hsa_circ_0000018) is a human DNAJC16-derived circRNA downregulated in LUAD tissues, with low expression associated with advanced stage, lymph node metastasis and poor overall survival. Functionally, circDNAJC16 suppresses LUAD proliferation, tumor growth, migration and invasion by sponging miR-93-5p and activating CDKN1A/p21-mediated G0/G1 arrest, while also inhibiting EMT and potentially Wnt/beta-catenin/STAT3 signaling. eIF4A3 binds upstream flanking regions of DNAJC16/circDNAJC16 and regulates circDNAJC16 subcellular localization, supporting its role as a prognostic biomarker and therapeutic target.

EIF4A3-mediated localization of circDNAJC16 sequesters miR-93-5p to suppress lung adenocarcinoma progression via CDKN1A-regulated cell cycle and EMT.
