circRNA-disease information
Expression pattern:
UP
Associated gene: LDHA mRNA, CFL1, AARS1
Associated microRNA: -
Biological function: Promotes NPC cell proliferation, invasion, migration, and metastasis by enhancing glycolysis and CFL1 lactylation; reduces cell stiffness and adhesion through cytoskeletal remodeling.
Molecular mechanism: circCDYL directly binds the 3'-UTR of LDHA mRNA to enhance LDHA mRNA stability, increasing LDHA protein levels and glycolysis; accumulated lactate induces AARS1-catalyzed CFL1 K22 lactylation, extending CFL1 half-life and promoting cytoskeletal remodeling.
Biological pathway or process:

glycolysis (promotes); proliferation (promotes); migration (promotes); invasion (promotes); metastasis (promotes); mRNA stability (promotes); other pathway/process (promotes)

Clinical significance:

circCDYL overexpression is significantly correlated with clinical staging and TNM staging of NPC and may serve as a potential therapeutic target.

Description:

circCDYL (hsa_circ_0008285), derived from CDYL exon 2, is overexpressed in nasopharyngeal carcinoma and correlates with clinical and TNM stage. It promotes NPC proliferation, invasion, migration, and metastasis by binding and stabilizing LDHA mRNA, increasing LDHA-driven glycolysis and lactate accumulation. Lactate then promotes AARS1-mediated CFL1 K22 lactylation, increasing CFL1 stability and cytoskeletal remodeling to reduce cell stiffness and adhesion.

Confidence score:

0.8826
