circRNA-disease information
Expression pattern:
UP
Associated gene: TRA2B, ELAVL1, 3'UTR of TRA2B, NF-kappaB2, NF-kappaB p65, p-NF-kappaB p65
Associated microRNA: miR-432-5p
Biological function: Promotes NSCLC cell migration and invasion in vitro, facilitates tumor growth in vivo, and enhances migratory and invasive capacities of recipient cells through exosomal delivery.
Molecular mechanism: Exosomal circ_0008803 acts through dual mechanisms: it sponges miR-432-5p to relieve repression of TRA2B, and binds ELAVL1 to promote ELAVL1 cytoplasmic translocation and stabilize TRA2B mRNA via 3'UTR interaction, thereby activating the NF-kappaB pathway.
Biological pathway or process:

NF-kappaB (promotes); migration (promotes); invasion (promotes); metastasis (promotes); proliferation (promotes); ceRNA regulation (promotes); mRNA stability (promotes); other pathway/process (promotes)

Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; Actinomycin D / DRB Stability Assay; RT-qPCR; Microarray; Clinical Sample Validation; RIP (RNA Immunoprecipitation); RNA Pull-Down; Luciferase Reporter Assay; Transfection; Wound Healing Assay; Transwell Assay; In Vivo Animal Model; Western Blot; IF (Immunofluorescence); Nuclear-Cytoplasmic Fractionation; ROC Analysis; Bioinformatics Analysis

Clinical significance:

Elevated serum exosomal circ_0008803 levels correlated with advanced NSCLC stages and distant metastasis, and serum exosomal circ_0008803 showed diagnostic potential for NSCLC.

Description:

circ_0008803 is an exosome-enriched RNF10-derived circRNA up-regulated in NSCLC serum exosomes and lung cancer cells. It promotes migration, invasion, metastasis-related phenotypes, and tumor growth by sponging miR-432-5p and binding ELAVL1 to increase TRA2B expression and activate NF-kappaB signaling. Serum exosomal circ_0008803 also shows diagnostic potential and correlates with metastasis and tumor stage.

Confidence score:

0.8791
