circRNA-disease information
Expression pattern:
DN
Associated gene: PI3K, AKT, mTOR
Associated microRNA: -
Biological function: CircSHPRH acts as a tumor suppressor in HNSCC by inhibiting cell proliferation, migration, invasion, and tumor growth, and by promoting apoptosis.
Molecular mechanism: CircSHPRH suppresses the PI3K/AKT/mTOR signaling pathway, reducing phosphorylation of PI3K, AKT, and mTOR; activation of PI3K by 740Y-P reverses circSHPRH-mediated phenotypes.
Biological pathway or process:

PI3K/AKT/mTOR (inhibits); proliferation (inhibits); migration (inhibits); invasion (inhibits); apoptosis (promotes); EMT (inhibits); metastasis (inhibits)

Detected method:
Q
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; Actinomycin D / DRB Stability Assay; RT-qPCR; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; Transfection; CCK8; Colony Formation Assay; Annexin V/PI Flow Cytometry; Wound Healing Assay; Transwell Assay; Western Blot; In Vivo Animal Model; IHC (Immunohistochemistry); TUNEL; Cohort Study; Survival Analysis

Clinical significance:

Low circSHPRH expression was associated with shorter overall survival and histological grade; circSHPRH may serve as a potential biomarker for HNSCC diagnosis, prognosis, and therapeutic response prediction.

Description:

CircSHPRH is a human circRNA derived from SHPRH and is significantly downregulated in HNSCC tissues and cell lines. In this study, circSHPRH overexpression suppressed proliferation, migration, invasion, EMT-related marker changes, and xenograft tumor growth while promoting apoptosis. Mechanistically, circSHPRH acted as a tumor suppressor at least partly by inhibiting PI3K/AKT/mTOR signaling, and higher expression was associated with better overall survival.
