circRNA-disease information
Expression pattern:
DN
Associated gene: GNAO1, DNMT1, RNA pol Ⅱ, Ago2
Associated microRNA: -
Biological function: Suppresses HCC cell proliferation, induces G1 cell cycle arrest, and promotes cellular senescence, thereby inhibiting HCC progression.
Molecular mechanism: Acts as an RBP sponge by sequestering DNMT1, reducing DNMT1 binding to the GNAO1 promoter and reversing DNMT1-induced promoter methylation, thereby enhancing GNAO1 transcription and increasing GNAO1 expression; not via miRNA sponge/ceRNA.
Biological pathway or process:

proliferation (inhibits); cell cycle (inhibits); senescence (promotes); m6A modification (not specified)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Survival Analysis; Bioinformatics Analysis; FISH / smFISH; Nuclear-Cytoplasmic Fractionation; Luciferase Reporter Assay; RNA Pull-Down; RIP (RNA Immunoprecipitation); ChIP / ChIP-seq; MSP (Methylation-Specific PCR); Transfection; CCK8; EdU Staining; Cell Cycle Assay; Flow Cytometry(Non-apoptosis/cycle); Western Blot

Clinical significance:

High expression of circGNAO1 was associated with high OS, while low expression of circGNAO1 was associated with low OS.

Description:

circGNAO1 (hsa_circ_0039422) is down-regulated in HCC tissues/cell lines and higher expression predicts better overall survival. Functionally, circGNAO1 suppresses HCC cell proliferation, induces G1 arrest, and promotes senescence by increasing its host gene GNAO1 transcription. Mechanistically, it binds/sequesters DNMT1 to reduce DNMT1 occupancy on the GNAO1 promoter and counteract DNMT1-driven promoter methylation.

Confidence score:

0.8133
