circRNA-disease information
Expression pattern:
DN
Associated gene: GADD45A, AGO2, p-JNK, Ki67
Associated microRNA: miR-331-5p
Biological function: Inhibits gastric cancer cell proliferation, migration, invasion, and subcutaneous tumor growth; acts as a tumor suppressor in GC.
Molecular mechanism: circDYRK1A functions as a competitive endogenous RNA by sponging miR-331-5p, relieving repression of GADD45A and activating the GADD45A-JNK axis.
Biological pathway or process:

proliferation (inhibits); migration (inhibits); invasion (inhibits); ceRNA regulation (promotes); MAPK (promotes)

Detected method:
Q
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; RT-qPCR; FISH / smFISH; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; RIP (RNA Immunoprecipitation); Luciferase Reporter Assay; Transfection; CCK8; EdU Staining; Transwell Assay; Western Blot; IHC (Immunohistochemistry); In Vivo Animal Model; Survival Analysis; Bioinformatics Analysis

Clinical significance:

Reduced circDYRK1A expression is associated with poor prognosis in GC patients; high circDYRK1A expression is associated with significantly higher overall survival.

Description:

circDYRK1A is a human DYRK1A-derived circRNA that is significantly downregulated in gastric cancer tissues and cell lines. It suppresses GC proliferation, migration, invasion, and xenograft tumor growth by sponging miR-331-5p, thereby restoring GADD45A expression and activating the GADD45A-JNK tumor-suppressive pathway. Clinically, higher circDYRK1A expression is associated with better overall survival in GC patients.

Confidence score:

0.8766
