circRNA-disease information
Expression pattern:
UP
Associated gene: HNF1beta, Derlin-1, Argonaute2
Associated microRNA: miR-217
Biological function: promotes cell proliferation, migration, and invasion; inhibits apoptosis; promotes glioma malignant progression
Molecular mechanism: Acts as a miR-217 sponge and forms an Argonaute2-dependent reciprocal negative feedback with miR-217, thereby regulating HNF1beta and downstream Derlin-1, involving PI3K/AKT and ERK signaling.
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); apoptosis (inhibits); PI3K/AKT (promotes); ERK (promotes); ceRNA regulation (promotes)

Detected method:
Q
H
Validation methods:

FISH / smFISH; RT-qPCR; RNase R Treatment; Transfection; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); CCK8; Transwell Assay; Annexin V/PI Flow Cytometry; Western Blot; In Vivo Animal Model; Survival Analysis; Clinical Sample Validation; Bioinformatics Analysis

Clinical significance:

Circ-TTBK2 expression was positively correlated with the pathological grades of glioma.

Description:

circ-TTBK2 is up-regulated in glioma tissues/cells and promotes malignant phenotypes (proliferation, migration, invasion) while inhibiting apoptosis. Mechanistically, it sponges miR-217 in an Ago2/RISC-dependent manner, thereby derepressing the miR-217 target HNF1beta and its downstream effector Derlin-1, with involvement of PI3K/AKT and ERK signaling and in vivo tumor growth effects.

Confidence score:

0.817
