circRNA-disease information
Expression pattern:
UP
Associated gene: MAPK14
Associated microRNA: miR-375
Biological function: circ_0008285 promotes LPS-induced macrophage inflammatory polarization, TC-1 cell injury and apoptosis, and lung injury in sepsis-induced ALI; knockdown of circ_0008285 reduces inflammatory cytokines, shifts macrophage markers toward M2 polarization, improves TC-1 viability, decreases apoptosis, and alleviates lung damage in CLP mice.
Molecular mechanism: circ_0008285 acts as a miR-375 sponge and regulates MAPK14 through the miR-375/MAPK14 axis, thereby affecting macrophage polarization, inflammatory injury and apoptosis in sepsis-induced ALI.
Biological pathway or process:

MAPK (promotes); ceRNA regulation (other); immune regulation (promotes); macrophage polarization (other); inflammation (promotes); apoptosis (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; Bioinformatics Analysis; Luciferase Reporter Assay; Transfection; Western Blot; CCK8; Annexin V/PI Flow Cytometry; In Vivo Animal Model; H&E Staining; TUNEL

Clinical significance:

Circ_0008285 may be served as a potential therapeutic object for sepsis-induced ALI.

Description:

This study found that circ_0008285 is up-regulated in LPS-treated macrophages and CLP-induced sepsis-ALI mouse lung tissue. Functionally, circ_0008285 promotes inflammatory macrophage polarization, epithelial cell injury/apoptosis, and lung damage, while its knockdown alleviates these phenotypes. Mechanistically, circ_0008285 acts through the miR-375/MAPK14 axis and may represent a therapeutic target for sepsis-induced ALI.

Confidence score:

0.5634
