circRNA-disease information
Expression pattern:
DN
Associated gene: HIF-1alpha, Ago2, caspase 3, BCL-2, BAX, cleaved-caspase 3, pro-caspase 3
Associated microRNA: miR-29a-3p
Biological function: circLRP6 inhibits AAA development and progression; circLRP6 overexpression inhibits Ang II-induced hAoSMC cell viability and proliferation, regulates cell cycle, and induces apoptosis.
Molecular mechanism: circLRP6 acts through the miR-29a-3p/HIF-1alpha axis, negatively regulates miR-29a-3p, interacts with HIF-1alpha, inhibits HIF-1alpha expression, and modulates apoptosis-related proteins.
Biological pathway or process:

HIF-1 signaling (inhibits); apoptosis (promotes); proliferation (inhibits); cell cycle (other); ceRNA regulation (other)

Detected method:

Validation methods:

RNase R Treatment; RT-qPCR; FISH / smFISH; Clinical Sample Validation; Bioinformatics Analysis; Transfection; CCK8; BrdU; Cell Cycle Assay; Annexin V/PI Flow Cytometry; Western Blot; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); IF (Immunofluorescence); In Vivo Animal Model

Clinical significance:

circLRP6 is proposed as a potential therapeutic target and theoretical basis for clinical optimization of AAA treatment.

Description:

circLRP6 is down-regulated in AAA tissues, Ang II-induced hAoSMCs, and mouse AAA aorta samples. Its overexpression suppresses Ang II-induced hAoSMC viability and proliferation, modulates cell cycle changes, and promotes apoptosis. Mechanistically, circLRP6 functions through a miR-29a-3p/HIF-1alpha-related axis and may serve as a potential therapeutic target for AAA.
