| Associated gene: |
TDP43, HDAC1, SERPINB2, uPA, uPAR |
| Associated microRNA: |

| Biological function: |
inhibits migration, invasion, vascular invasion, and metastasis of HCC cells |
| Molecular mechanism: |
circPSD3 interacts with HDAC1 and sequesters it in the cytoplasm, attenuating HDAC1-mediated transcriptional repression of SERPINB2 and inhibiting the uPA system; TDP43 suppresses circPSD3 biogenesis |
| Biological pathway or process: |
migration (inhibits); invasion (inhibits); metastasis (inhibits); other pathway/process (inhibits) |

circRNA-seq; RNase R Treatment; RT-qPCR; Sanger Sequencing; Back-Splice Junction PCR / divergent primers PCR; Actinomycin D / DRB Stability Assay; Nuclear-Cytoplasmic Fractionation; FISH / smFISH; RIP (RNA Immunoprecipitation); Transfection; Western Blot; IHC (Immunohistochemistry); IF (Immunofluorescence); CCK8; Colony Formation Assay; Wound Healing Assay; Transwell Assay; Luciferase Reporter Assay; In Vivo Animal Model; H&E Staining; Bioinformatics Analysis; Survival Analysis |
| Clinical significance: |
Decreased circPSD3 expression in HCC tissues predicted poor prognosis (recurrence-free survival and overall survival) and was associated with unfavourable characteristics including MVI. |
| Description: |
circPSD3 (hsa_circ_0136098), derived from PSD3, is down-regulated in PVTT and HCC tissues and its low expression predicts worse survival. Functionally, circPSD3 suppresses HCC cell migration/invasion and inhibits vascular invasion and metastasis in vivo by binding HDAC1 to retain it in the cytoplasm, thereby relieving repression of SERPINB2 and inhibiting the uPA/uPAR system. |
