circRNA-disease information
Expression pattern:
UP
Associated gene: TLR-4, NF-kappaB, TLR4, IkappaBalpha, p65
Associated microRNA: miR-326
Biological function: Circ_0001715 promotes proliferation and inflammation and suppresses apoptosis in RA fibroblast-like synoviocytes; knockdown of circ_0001715 decreases proliferation and inflammation, promotes apoptosis, and mitigates pathological manifestations in the CIA mouse model.
Molecular mechanism: Circ_0001715 acts through a ceRNA mechanism by sequestering miR-326, thereby regulating the TLR-4/NF-kappaB axis in RA fibroblast-like synoviocytes.
Biological pathway or process:

NF-kappaB (promotes); proliferation (promotes); apoptosis (inhibits); inflammation (promotes); ceRNA regulation (other)

Detected method:
Q
M
Validation methods:

RT-qPCR; Microarray; Clinical Sample Validation; Luciferase Reporter Assay; Transfection; CCK8; Annexin V/PI Flow Cytometry; ELISA; Western Blot; In Vivo Animal Model; H&E Staining; Bioinformatics Analysis

Clinical significance:

Circ_0001715 may be a target for the diagnosis and treatment of RA.

Description:

Circ_0001715 is up-regulated in RA patient synovial tissues, TNF-alpha-induced HFLS-RA cells, and CIA mouse synovial tissues. Functionally, circ_0001715 promotes proliferation and inflammation and inhibits apoptosis in RA-FLSs. Mechanistically, it sequesters miR-326 to regulate the TLR-4/NF-kappaB axis, suggesting potential diagnostic and therapeutic value in RA.

Confidence score:

0.6835
