circRNA-disease information
Expression pattern:
UP
Associated gene: P65, P50, NF-kappaB, KDM5A, SOCS1, VEGFR3, TET1, AGO2
Associated microRNA: miR-4711-5p
Biological function: circDMD promotes tumor proliferation, metastasis, autophagy, migration, invasion, angiogenesis-like tube formation, and tumor growth in cervical cancer models.
Molecular mechanism: circDMD is induced by TNFalpha, interacts with P65/P50, promotes NF-kappaB expression and nuclear translocation, sponges miR-4711-5p to increase KDM5A and reduce SOCS1-mediated P65 ubiquitination, and promotes VEGFR3 expression by forming an R-loop at the VEGFR3 promoter.
Biological pathway or process:

NF-kappaB (promotes); proliferation (promotes); migration (promotes); invasion (promotes); metastasis (promotes); autophagy (promotes); angiogenesis (promotes); ubiquitination (inhibits); ceRNA regulation (promotes); other pathway/process (promotes)

Detected method:

Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; Actinomycin D / DRB Stability Assay; RT-qPCR; RNA-seq; FISH / smFISH; RIP (RNA Immunoprecipitation); RNA Pull-Down; Co-IP; Luciferase Reporter Assay; Transfection; MTT; Transwell Assay; Tube Formation Assay; IF (Immunofluorescence); In Vivo Animal Model; IHC (Immunohistochemistry); Western Blot; Bioinformatics Analysis

Clinical significance:

circDMD may provide a new potential biomarker for cervical cancer.

Description:

circDMD is a TNFalpha-induced circRNA derived from DMD that promotes cervical cancer tumorigenesis. It activates NF-kappaB signaling through P65/P50 interaction, a miR-4711-5p/KDM5A/SOCS1 ubiquitination-related mechanism, and R-loop formation at the VEGFR3 promoter, thereby enhancing proliferation, metastasis, autophagy, angiogenesis, and xenograft tumor growth.
