circRNA-disease information
Expression pattern:
UP
Associated gene: TGF-alpha
Associated microRNA: miR-144-3p
Biological function: Promotes cisplatin resistance, enhances autophagy, increases cell viability/proliferation, inhibits apoptosis, and promotes tumor growth in thyroid carcinoma cells.
Molecular mechanism: Acts as a miRNA sponge for miR-144-3p, releasing repression of TGF-alpha and thereby activating autophagy under cisplatin treatment, leading to reduced apoptosis and increased cisplatin resistance.
Biological pathway or process:

autophagy (promotes); apoptosis (inhibits); proliferation (promotes); chemoresistance (promotes); ceRNA regulation (promotes)

Detected method:
Q
M
Validation methods:

Microarray; Bioinformatics Analysis; RT-qPCR; Clinical Sample Validation; RNA Pull-Down; Luciferase Reporter Assay; Transfection; Western Blot; IF (Immunofluorescence); IHC (Immunohistochemistry); MTT; Annexin V/PI Flow Cytometry; In Vivo Animal Model

Clinical significance:

Description:

This study reports that circEIF6 (hsa_circ_0060060) is upregulated in thyroid carcinoma and promotes cisplatin resistance. Mechanistically, circEIF6 sponges miR-144-3p to increase TGF-alpha expression, thereby enhancing autophagy and inhibiting apoptosis in PTC/ATC cells, and circEIF6 knockdown increases cisplatin sensitivity in a BHT101 xenograft model.

Confidence score:

0.7756
