circRNA-disease information
Expression pattern:
UP
Associated gene: METTL14, MAP3K9, Ago2
Associated microRNA: miR-186-3p
Biological function: Promotes GC cell growth, colony formation, invasion, metastasis, tumorigenesis and cisplatin/DDP resistance; circUGGT2 knockdown suppresses these malignant phenotypes.
Molecular mechanism: METTL14-dependent m6A modification downregulates circUGGT2; circUGGT2 acts as a miR-186-3p sponge to upregulate MAP3K9, thereby promoting GC progression and DDP resistance.
Biological pathway or process:

m6A modification (other); ceRNA regulation (promotes); proliferation (promotes); invasion (promotes); metastasis (promotes); chemoresistance (promotes)

Detected method:
Q
H
Validation methods:

RNase R Treatment; RT-qPCR; Microarray; MeRIP / MeRIP-seq; RIP (RNA Immunoprecipitation); FISH / smFISH; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; Luciferase Reporter Assay; Bioinformatics Analysis; Transfection; MTT; Colony Formation Assay; Transwell Assay; Western Blot; In Vivo Animal Model; H&E Staining; IHC (Immunohistochemistry); Survival Analysis

Clinical significance:

Upregulated circUGGT2 is associated with poor overall survival in GC patients and may provide a prognostic and therapeutic target for GC.

Description:

circUGGT2 is an upregulated oncogenic circRNA in gastric cancer and cisplatin-resistant GC cells. METTL14-dependent m6A modification reduces circUGGT2 expression, while circUGGT2 promotes tumor growth, invasion, metastasis and DDP resistance by sponging miR-186-3p and upregulating MAP3K9. High circUGGT2 expression is associated with poor overall survival in GC patients.

Confidence score:

0.8583

Other information
Title:

CircUGGT2 downregulation by METTL14-dependent m6A modification suppresses gastric cancer progression and cisplatin resistance through interaction with miR-186-3p/MAP3K9 axis.
