circRNA-disease information
Expression pattern:
UP
Associated gene: KIF3A, kinesin family member 3A, PCNA, MMP-2, MMP-9
Associated microRNA: miR-582-3p
Biological function: Promotes bladder cancer cell proliferation, migration, invasion, metastasis-related phenotypes, and xenograft tumor growth; knockdown inhibits these malignant phenotypes.
Molecular mechanism: ceRNA mechanism: circRAPGEF5 sponges miR-582-3p to increase KIF3A expression, thereby promoting malignant phenotypes of bladder cancer cells.
Biological pathway or process:

ceRNA regulation (promotes); proliferation (promotes); migration (promotes); invasion (promotes); metastasis (promotes)

Detected method:
Q
Validation methods:

RNase R Treatment; RT-qPCR; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; Transfection; CCK8; Colony Formation Assay; Wound Healing Assay; Transwell Assay; Western Blot; Bioinformatics Analysis; Luciferase Reporter Assay; RNA Pull-Down; In Vivo Animal Model; H&E Staining; Survival Analysis

Clinical significance:

High circRAPGEF5 expression is associated with lower overall survival and is closely related to tumor size and tumor stage in bladder cancer patients; it may serve as a potential biomarker and therapeutic target.

Description:

circRAPGEF5 is up-regulated in bladder cancer tissues and cell lines, and high expression is associated with poor overall survival, tumor size, and tumor stage. Functionally, circRAPGEF5 promotes BCa cell proliferation, migration, invasion, and xenograft tumor growth. Mechanistically, it acts as a ceRNA by sponging miR-582-3p and up-regulating KIF3A.

Confidence score:

0.8175
