circRNA-disease information
Expression pattern:
DN
Associated gene: FTO, UBC13
Associated microRNA: -
Biological function: promotes vascular repair; promotes vessel regeneration and perfusion; repairs BBB integrity following stroke
Molecular mechanism: circSCMH1 binds FTO and promotes its nuclear translocation via UBC13-mediated Ub-K63 ubiquitination, leading to m6A demethylation of Plpp3 mRNA and increased Plpp3/LPP3 expression (enhanced mRNA stability).
Biological pathway or process:

m6A modification (inhibits); mRNA stability (promotes); angiogenesis (promotes); other pathway/process (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; RNA Pull-Down; RIP (RNA Immunoprecipitation); Western Blot; Transfection; In Vivo Animal Model; IF (Immunofluorescence); Flow Cytometry(Non-apoptosis/cycle); MeRIP / MeRIP-seq; Actinomycin D / DRB Stability Assay; Bioinformatics Analysis

Clinical significance:

circSCMH1 is decreased in somatosensory cortex of AIS patients versus nonstroke controls.

Description:

circSCMH1 is down-regulated after ischemic stroke and, when supplemented (EV-delivered/overexpressed), promotes vascular repair and BBB restoration. Mechanistically, it binds FTO and enhances UBC13-dependent Ub-K63 ubiquitination and nuclear translocation of FTO, reducing m6A on Plpp3 mRNA, increasing Plpp3/LPP3 expression via improved mRNA stability, and thereby improving stroke recovery.

Confidence score:

0.7198
