circRNA-disease information
Expression pattern:
UP
Associated gene: HuR, ZO-1, ZO-1 mRNA
Associated microRNA: -
Biological function: Promotes proliferation, migration, invasion/metastasis and EMT progression of HCC cells; exosomal circ-0004277 stimulates EMT and migration of surrounding normal hepatic cells.
Molecular mechanism: circ-0004277 competitively binds the RBP HuR, blocks HuR binding to ZO-1 mRNA, inhibits ZO-1, and thereby stimulates EMT progression.
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); metastasis (promotes); EMT (promotes); other pathway/process (inhibits)

Detected method:
Q
Validation methods:

RT-qPCR; RNase R Treatment; Sanger Sequencing; Clinical Sample Validation; ROC Analysis; Transfection; CCK8; EdU Staining; Transwell Assay; Wound Healing Assay; RIP (RNA Immunoprecipitation); Western Blot; IF (Immunofluorescence); IHC (Immunohistochemistry); In Vivo Animal Model; H&E Staining; Bioinformatics Analysis

Clinical significance:

Exosomal circ-0004277 may be a diagnostic biomarker for distinguishing HCC patients from negative controls (ROC AUC = 0.816).

Description:

hsa_circ_0004277 (circ-0004277), derived from WDR37, is up-regulated in HCC tissues, HCC cell lines, and plasma exosomes, and promotes HCC proliferation, migration, EMT, and metastasis. Mechanistically, it binds the RBP HuR, interferes with HuR-ZO-1 mRNA interaction, suppresses ZO-1, and thereby enhances EMT. Exosomal circ-0004277 can be transferred to surrounding normal hepatic cells to stimulate EMT/migration, and it shows diagnostic potential by ROC analysis.

Confidence score:

0.8202
