circRNA-disease information
Expression pattern:
UP
Associated gene: EGFR, AGO2
Associated microRNA: miR-491-5p
Biological function: promotes proliferation, cell cycle progression, migration, invasion, tumor growth/metastasis; promotes immunosuppression and anti-PD-1 therapy resistance
Molecular mechanism: ceRNA/miRNA sponge: circ_CELF1 binds miR-491-5p to increase EGFR expression, contributing to NSCLC progression and immunotherapy resistance
Biological pathway or process:

proliferation (promotes); cell cycle (promotes); migration (promotes); invasion (promotes); metastasis (promotes); immune regulation (inhibits); other pathway/process (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; FISH / smFISH; RIP (RNA Immunoprecipitation); Luciferase Reporter Assay; Transfection; CCK8; Colony Formation Assay; Wound Healing Assay; Transwell Assay; Cell Cycle Assay; Western Blot; IHC (Immunohistochemistry); ROC Analysis; Survival Analysis; In Vivo Animal Model; H&E Staining; Bioinformatics Analysis

Clinical significance:

High level of circ_CELF1 is correlated with poor prognosis and elevated recurrence; ROC suggests diagnostic value in tissues (AUC reported in figure).

Description:

circ_CELF1 is up-regulated in NSCLC tissues and promotes malignant phenotypes (proliferation, cell-cycle progression, migration/invasion, tumor growth/metastasis). It functions mainly as a ceRNA by sponging miR-491-5p to upregulate EGFR, and is associated with reduced CD8+ T-cell infiltration and resistance to anti-PD-1 therapy.

Confidence score:

0.8596
