circRNA-disease information
Expression pattern:
UP
Associated gene: EEF1A2, COL14A1, COL14A1 promoter, CDYL, EIF4A3, HIF-1⍺
Associated microRNA: -
Biological function: Promotes ICAM-1+ CSC/CCSC generation, epithelial-mesenchymal transition, bloodstream entry, and lung metastasis in HCC.
Molecular mechanism: Hypoxia-induced HIF-1⍺ upregulates CDYL and EIF4A3 to increase circ-CDYL; nuclear circ-CDYL cooperates with the RNA-binding protein EEF1A2 to activate COL14A1 transcription, enhancing ERK signaling and epithelial-mesenchymal transition.
Biological pathway or process:

HIF-1 signaling (other); ERK (promotes); EMT (promotes); metastasis (promotes); ceRNA regulation (other); other pathway/process (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; FISH / smFISH; Nuclear-Cytoplasmic Fractionation; RIP (RNA Immunoprecipitation); RNA Pull-Down; ChIP / ChIP-seq; Transfection; Flow Cytometry(Non-apoptosis/cycle); In Vivo Animal Model; H&E Staining; Western Blot; Bioinformatics Analysis; Digital PCR

Clinical significance:

circ-CDYL is elevated in HCC patient CCSCs and liver CSCs; the circ-CDYL-related ICAM-1+ CCSC system may support preoperative and postoperative HCC treatment decisions related to lung metastasis risk.

Description:

In HCC, circ-CDYL is up-regulated in ICAM-1+ CCSCs and liver CSCs and promotes lung metastasis. Mechanistically, hypoxia-induced HIF-1⍺ increases circ-CDYL through CDYL and EIF4A3, while nuclear circ-CDYL cooperates with EEF1A2 to activate COL14A1 transcription, leading to ERK signaling activation and EMT.

Confidence score:

0.7663
