circRNA-disease information
Expression pattern:
UP
Associated gene: EZH2, AGO2, Wnt10b, Wnt1
Associated microRNA: miR-138-5p
Biological function: promotes preadipocyte differentiation/adipogenesis and is associated with obesity prognosis; knockdown reduces differentiation and improves obesity-related phenotypes in vivo
Molecular mechanism: Acts as a miRNA sponge for miR-138-5p to increase EZH2 expression (ceRNA-like regulation), involving AGO2/RISC association
Biological pathway or process:

ceRNA regulation (promotes); proliferation (other)

Detected method:
Q
H
M
Validation methods:

Microarray; RNase R Treatment; RT-qPCR; divergent primers PCR; Sanger Sequencing; Actinomycin D / DRB Stability Assay; FISH / smFISH; ISH (In Situ Hybridization); RIP (RNA Immunoprecipitation); circRIP; RNA Pull-Down; Luciferase Reporter Assay; Transfection; Oil Red O Staining; Western Blot; IHC (Immunohistochemistry); In Vivo Animal Model; H&E Staining; ROC Analysis; Cohort Study; Bioinformatics Analysis

Clinical significance:

circSAMD4A upregulation in adipose tissues was correlated with a poor prognosis in obese patients and may predict non-remission after bariatric surgery

Description:

circSAMD4A (hsa_circ_0004846), derived from hSAMD4A, is up-regulated in visceral adipose tissue of obese patients and predominantly localized in the cytoplasm. It promotes preadipocyte differentiation by sponging miR-138-5p, thereby relieving repression of EZH2 and supporting an circSAMD4A/miR-138-5p/EZH2 regulatory axis. High circSAMD4A expression is associated with poor prognosis (non-remission) after bariatric surgery and shows diagnostic performance by ROC analysis.
